Introduction

In the early 1970s, researchers and clinicians began to investigate whether and to what extent quetiapine (Seroquel, Seroquel, Seroquel) could treat bipolar disorder (BD) in adults [

]. By 1975, researchers began to investigate the efficacy and safety of this medication. In particular, a large-scale clinical trial (STR) with quetiapine was conducted and its findings were significant to determine the optimal dosing regimen [

This trial included 18 participants with BD, who were randomized to receive either 100 mg or 200 mg quetiapine daily for 5 days, or placebo. The researchers evaluated the efficacy, tolerability, and safety of the quetiapine group, the placebo group, and the combination group. The researchers determined that the quetiapine group showed improved efficacy, improved tolerability, and a better safety profile.

Quetiapine was first approved by the Food and Drug Administration (FDA) in 1985 and has since been used for several conditions including major depressive disorder [

It was also approved by the FDA in 1987 and has been used for the treatment of attention-deficit/hyperactivity disorder (ADHD) [

The study also demonstrated that the use of quetiapine can improve the treatment-emergent (TEE) response to TEE (1–2 weeks) in bipolar disorder patients [

Since its approval, quetiapine has been used to treat multiple conditions, including depression, mania, schizophrenia, obsessive-compulsive disorder (OCD), and bipolar disorder. Its use in these conditions has also been supported by numerous clinical trials and the results have shown an increased treatment response, with patients reporting a decrease in depression [

Quetiapine has been used to treat bipolar disorder, schizophrenia, OCD, and major depressive disorder, but the evidence for its efficacy in these conditions has been inconclusive [

Therefore, the study included participants with BD who were randomly assigned to receive either quetiapine or placebo at week 1, week 2, and week 4 after randomization. The researchers then assessed the safety of the study, the treatment-emergent (TEE) response, and the TEE response in these participants, and conducted a safety analysis to determine the optimal dosing regimen. Both of these studies included participants with BD who were randomized to either quetiapine or placebo, and the safety of this group was determined.

Quetiapine is a well-known and well-studied medication, and it has been approved by the FDA and the European Medicines Agency (EMA) for the treatment of bipolar I disorder. The EMA also approved quetiapine for the treatment of major depressive disorder and anxiety disorders [

Quetiapine is aripiprazole, a proton pump inhibitor (PPI) that is FDA-approved for the treatment of BD and has been used for several conditions such as major depressive disorder, mania, schizophrenia, OCD, and bipolar disorder [

In addition to its FDA-approved uses, quetiapine has been used to treat both manic and depressive episodes in adults with major depressive disorder and anxiety disorders [

Quetiapine is a selective serotonin reuptake inhibitor (SSRI) used to increase serotonin levels in the brain, helping to improve mood and reduce symptoms of depression and anxiety. Quetiapine works by increasing the levels of serotonin in the brain, helping to improve feelings of pleasure, frustration and sadness. Quetiapine is available in tablet and capsule form and is usually taken once daily at a dosage of 300 mg or twice daily, depending on the severity of the disorder. Quetiapine is not recommended for children under the age of 18 years as it may cause problems in the development of the brain and may affect the ability to make mood-related decisions.

Background:Clinical practice guidelines recommend quetiapine as first-line treatment for depression, but only for patients with comorbid depressive symptoms or those with severe co-occurring disorders. Randomized clinical trials have been performed to investigate the efficacy of quetiapine in combination with other treatments for the treatment of depression.

Objectives:Quetiapine was compared to placebo in patients with depressive disorder and schizophrenia with or without major depressive episodes.

Study Design:Randomised double-blind placebo-controlled double-dummy controlled trial.

Participants:Patients with schizophrenia who responded to the first study visit of the schizophrenia treatment (DSM-5) and who responded to the second study visit of the schizophrenia treatment (SSRIs) were excluded from the study.

Patients who did not respond to the first study visit of the schizophrenia treatment, who responded to the second study visit of the schizophrenia treatment and who responded to the third study visit of the schizophrenia treatment.

Main outcome measures:Generalised anxiety disorder (GAD), panic disorder (PD), major depressive disorder (MDD), schizophrenia, bipolar disorder, depressive episode, and treatment response.

Statistical analysis:The primary outcome measure was response to the treatment group (placebo) or the second treatment group (Quetiapine). The secondary outcomes were treatment response (in terms of change in clinical symptoms and/or response to the treatment) and treatment symptom severity.

Results:The trial was primarily designed to test the effectiveness of Quetiapine (Seroquel®) in the treatment of patients with schizophrenia. Quetiapine was well tolerated in most of the included patients. However, there was a significant difference in the response rates between the quetiapine and placebo groups. Patients treated with Quetiapine (n = 26) reported significantly greater improvement in depressive symptoms in addition to their clinical symptoms. This finding was corroborated by a significant increase in response rates for both quetiapine and placebo treatment groups (p = 0.021 and p = 0.001, respectively).

Conclusions:Quetiapine is an effective treatment option for patients with schizophrenia and a well-tolerated treatment for depression.

Quetiapine (Seroquel®) is not a new drug. It was first developed in the late 1970s as an antipsychotic drug to treat bipolar disorder. Quetiapine was originally used to treat schizophrenia in bipolar disorder by the French psychiatrist Charles Bellet (1849–1918). In 1990, it was approved as an antipsychotic drug for treatment of bipolar disorder. Quetiapine is not currently indicated for the treatment of bipolar disorder. It is currently unavailable. Quetiapine has been shown to be an effective treatment option for patients with major depressive disorder and is considered to be safe and well tolerated. Patients with major depressive disorder and schizophrenia are at an increased risk of suicide and are more likely to be suicidal (, ). In addition, quetiapine is associated with an increased risk of suicide.

In clinical practice, quetiapine is used to treat bipolar depression. The mechanism of action for quetiapine in bipolar depression is unknown. However, a review of the literature suggests that it may be effective in patients with major depressive disorder and schizophrenia. Quetiapine has been shown to be a safe and effective treatment option for depression, especially in patients with depressive symptoms that were not responding to antidepressant therapy.

A clinical practice guideline (the National Institute for Health and Care Excellence) recommended quetiapine as the first-line therapy for patients with major depressive disorder and schizophrenia in adults with depression and bipolar disorder (, ). The guideline recommended quetiapine for treatment of major depressive disorder in adults with both depression and bipolar disorder. In the treatment of bipolar disorder in adults with depression, quetiapine is generally considered safe and well tolerated. However, the evidence is mixed and there is insufficient evidence to recommend it as a treatment for schizophrenia. Quetiapine is not indicated for the treatment of bipolar disorder in adults with bipolar disorder.

Quetiapine is not currently indicated for the treatment of patients with schizophrenia.

Quetiapine has been approved for the treatment of patients with depression. The Food and Drug Administration (FDA) approved quetiapine as an add-on treatment for adults with depression in the United States (U. S.) (, ).
